Relation locus of control on prevalence of depresivity by infertil person

This master thesis deals with infertility and its treatment. Both in the theoretical and empirical part, it focuses on the female aspects of the issue. The theoretical part summarizes an overview of key information important to understand the presented problems. We define the terms, describe the psychological and physiological causes of infertility, the process of treatment of infertility and aspects thereof. We focus on the depression rate related to infertility and its treatment, furthermore, we deal with type locus of control related to the experienced rate of depression. Our aim in the empirical part is to describe the depression rate, type of locus of control within the two samples of women acquired by us and capture the possible connections. The sample is made of 58 women who are currently undergoing a treatment for infertility and of the control group, which is made of 58 healthy women. To collect the data, we used the Beck Depression Inventory BDI-II, the Rotter's Internal- External Scale and the anamnestic questionnaire. Based on acquired data, we found out some specifity in depression rate and type of locus of control within our samples of women.
Rab proteins in sperm life

Rab proteins are the largest group of small GTPases. They work as key regulators in the intracellular vesicular transport of cells. This bachelor's thesis contains a literature review of current information on the function of Rab proteins in mammalian spermatozoa. The first part of the work is devoted to their structure, function, cycle, and diseases related to the dysfunction of Rab proteins. The second part is focused on sperm development and maturation linked with specific Rab proteins involved in individual steps during the life of the sperm. Rab proteins are important in cytoskeletal organization and cytokinesis of sperm during spermatogenesis. They participate in the acrosome biogenesis mediated by the Golgi apparatus and the morphological changes of sperm during spermiogenesis. They also play a key role in the acrosome reaction and interaction with the zona pellucida. Due to their necessity in the sperm development and maturation, Rab proteins are proposed as a marker of male fertility, which is summarized in the last part of the thesis. Therapeutic influence of Sertoli cells on the effects of oxidative stress in germ cells

Sertoli cells are somatic cells found in the testes and have a function in supporting spermatogenesis and germ cell nutrition, but also in maintaining an anti-inflammatory environment and forming the blood-testicular barrier in the testes. They are intensively researched for their immunomodulating properties which can be used in transplant medicine or as a therapeutic tool in regenerative medicine. Following previous research confirming that mouse Sertoli cells share some characteristics of mesenchymal stem cells and have multipotent differentiation potential in vitro, experiments were conducted to determine their therapeutic capabilities. Sertoli cells have demonstrated the ability to promote the maintenance of sperm motility during testicular inflammation and reduce the amount of immotile sperm in vivo. Based on preliminary results, they can also reduce the amount of ROS produced by testicular cells affected by oxidative stress in vitro. Ethical issues of surrogacy

This thesis has a theoretical character and deals with the issue of surrogacy in the Czech Republic. The aim of this thesis is to approach the essential ethical issues and show how the institute of surrogacy is regulated by Czech law. Furthermore, this work shows the role played by a social worker in the institute and it also deals with the family, which is unique and an irreplaceable institution. It is known that the number of infertile couples is increasing today and surrogacy is one of the methods of infertility assistance in which even these couples can become parents. In the Czech Republic, however, the institute of surrogacy is only minimally regulated by law and is ethically unacceptable to many people around the world. At this thesis, I point out that it is important that, in future, the institute of surrogacy be fully regulated, that there should be no abuse of this institute and that the negative effects that may occur should be avoided.
In vitro fertilization from the point of view of the couple

This bachelor thesis focuses on handeling and perception of assisted reproduction by a couple. The theoretical part provides information from the field of infertility, its examination and treatment options and especially the psychological aspects that appear with infertility treatment. The research study is based on qualitative research in which was used semi-structured interviews with six respondents who experienced assisted reproduction with successful conclusion. These respondents were asked about their subjective experience with the whole process and how this experience influenced their lives. The results of the thesis include individual case studies that describe the unique stories of the respondents. The data were analyzed using open coding and processed in the form of case studies. The results show that the respondents consider the process of assisted reproduction as a psychologically demanding which affects not only themselves and their lives but also affects relationship with the partner and external environment.
Ethical aspects of assisted reproduction as a challenge to the social worker´s action

The thesis deals with the issue of assisted reproduction, the definition of methods and its ethical issues. It defines infertility as a medical but also social problem. It focuses on the principles of social work and describes its ethical nature. In this way, it links the medical problem with the social problem and emphasizes the need for multidisciplinary cooperation. The thesis draws attention to the human being as a patient in the process of assisted reproduction, compares the current situation in the Czech Republic with selected foreign practices and defines the possibilities of the social worker in health care and its importance. The work is shaped by theoretical background with emphasis on current health statistics supplemented by sources of foreign practice.
Ethical aspects of dignified early life

The thesis focuses on ethical aspects influencing dignified beginning of life. Main subject is the issue of infertility and possible ways of its resolution. Introduction part defines concept of infertility, analyzes causes of infertility and presents the most common methods of assisted reproduction. Fundamental ethical problems associated with individual methods of assisted reproduction are analyzed in next section. Attention is also paid to legislation of assisted reproduction in the Czech Republic, not only to legal framework of individual methods but also to determination of motherhood and paternity. Separate chapter is devoted to issue of surrogate motherhood which is a current and highly controversial topic. Conclusion of the thesis is dedicated to psychological impacts on couples living together and opportunities for social work.

Assisted reproduction - legal and ethical aspects Abstract This master's thesis discusses legal and ethical aspects of assisted reproduction. The aim of this thesis is to describe current legislation governing assisted reproduction, evaluate whether such legislation is able to meet the needs of modern society and also to discuss possible amendments thereof necessary for establishing a reasonable and effective legal system. With respect to the fact that assisted reproduction still represents a controversial topic among the general public, this thesis also deals with related ethical aspects and mentions the most contentious topics debated both in the Czech Republic and worldwide. This thesis is divided into six chapters. The first chapter explains the term assisted reproduction and describes its main principles, methods and procedures along with conditions for assisted reproduction including infertility. The second chapter of this thesis is more practical and focuses on topics related to providing assisted reproduction, such as who may provide said medical service or who may be the recipient thereof. The mechanism of impaired formation of head-tail coupling apparatus in centrobin deficient acephalic spermatozoa

Acephalic sperm syndrome (ASS) is a rare form of teratozoospermia that is probably genetic in origin. The sperm of individuals with this syndrome have a damaged apparatus connecting the head and flagellum (HTCA), which leads to the formation of acephalic sperm, i.e. sperm with separate heads from flagella. Individuals affected by ASS are almost exclusively infertile. One of the proteins whose mutation causes ASS is SUN5. Together with KASH proteins, SUN proteins are part of the so-called LINC complex, which ensures the connection of the nucleoskeleton with the cytoskeleton. Centrobin is a protein involved in centriole duplication and assembly of the dividing spindle. Rats carrying a defective gene for centrobin have reduced fertility and exhibit an ASS phenotype. For that reason, this model organism was used in this work to study the possible interrelationships of proteins involved in the formation of ASS. Given the similar phenotype of centrobin and SUN5 mutated spermatozoa, there is a possibility that centrobin interacts with one of the LINC complex proteins and together they participate in the formation of the head-flagella junction.
